A master’s thesis was discussed at the College of Science, University of Misan, tagged “Biochemical and Molecular study of Bacteria Isolated from Frozen Food in local market in Misan Governorate” which was submitted by student (Doaa Ali Hussein ) and supervised by Prof. Dr. Zahid Saadoun Aziz.
The thesis reported the collection of 100 different samples of different products including red meat and chicken from local and foreign imported companies. The study aimed to know the bacterial load of the samples, and isolation and diagnosing of different types of Gram-positive and Gram-negative bacteria by traditional culture methods and the Phytec 2 device, as well as to conduct molecular diagnosis by using special and general primers and by the technique of single and multiple polymerase chain reaction. The DNA of studied samples were sequenced and 16 bacterial species were recorded in the gene bank under various regular numbers.
The study indicated that there is very high pollution in the samples under study, and that the percentage of contamination was higher in imported meat and chicken products compared to local ones. The study concluded that bacterial contamination is high in frozen meat and need more techniques to improve meat quality and follow up on its sources and methods of manufacture and storage to be suitable for human consumption. The study also concluded that molecular methods were better in diagnosis than using Phytic 2 device.
The study recommended tightening the role of health control over imported meat at border crossings, as well as its storage at consumer sales outlets.
The thesis was awarded with excellent grade.
